RAD-140 (Testolone):
What You NEED To Know

What is RAD-140?

RAD-140, also known as Testolone, belongs to the class of selective androgen receptor modulators (SARMs). It mimics testosterone's anabolic effect and improves muscle growth and fat loss.

RAD-140 acts more directly (more selectively) on the androgen receptor than testosterone and therefore has fewer side effects. The SARM is currently being researched in phase 1/2 clinical trials in humans. In the future, it could be used as a drug against muscle atrophy and breast cancer.

Benefits of RAD-140

  • Rapid muscle growth: RAD-140 binds to the androgen receptor, triggering various anabolic processes in the body, including improved muscle growth.
  • Fat loss: RAD-140 promotes fat burning by increasing metabolic rate and thus calorie consumption.
  • Neuroprotective effects: Some studies suggest that RAD-140 may protect brain cells.
  • Selective effect: As a SARM, RAD-140 acts selectively on the androgen receptor and, therefore, has fewer side effects.

How Does RAD-140 Work?

RAD-140 is a selective androgen receptor modulator (SARM). After ingestion, it binds to the androgen receptors (AR) present in many areas of the body. These receptors can be found in muscles, bones, or the prostate.

By selectively binding to the androgen receptors, the receptors change their structure, which enables it to interact with other molecules in the cell. These interactions then lead to the actual anabolic effect of RAD-140: It promotes muscle growth, accelerates recovery, and reduces muscle breakdown.

RAD-140 usually has fewer side effects than the hormone testosterone, which also gets its anabolic effect by binding to the androgen receptor. This is because testosterone is equally active in all tissues that have androgen receptors. RAD-140 has a more tissue-specific effect and largely avoids activating the receptors in the prostate or hair follicles. This has the advantage that side effects such as prostate enlargement or hair loss are less likely with RAD-140.

Side Effects of RAD-140 (Testolone)

Common mild side effects:

  • Headaches
  • Nausea
  • Acne

Severe, rare side effects:

  • Can cause gyno in rare cases
  • Suppresses testosterone production
  • Liver toxicity

Does RAD-140 damage the liver?

Yes, RAD-140 could potentially damage the liver. In 2022, a study was published in which a 24-year-old man suffered from abdominal pain, jaundice, and other complaints after taking RAD-140 for 5 weeks. A liver biopsy showed RAD-140-associated liver injury [R]. However, liver damage appears to be very rare. To date, there are only a few studies that suggest a connection between liver toxicity and RAD-140.

Does RAD-140 (Testolone) suppress testosterone?

Yes, RAD-140 (Testolone) can suppress the body's production of testosterone. This is not because RAD-140 increases testosterone levels in the body, because it does not. Rather, it is due to the binding to the androgen receptor. This can lead to negative feedback, which can reduce the body's production of testosterone.

The body regulates the production of the hormone testosterone through the hypothalamic-pituitary-gonadal axis (HHG axis). The androgen activity in the body is constantly "measured" by the hypothalamus. This activity can be significantly increased by a SARM such as RAD-140 because it binds to precisely these androgen receptors. When the hypothalamus measures an increase in androgen activity, testosterone production in the testes is reduced through a complex process.

RAD-140 Before and After

A user completed a 7-week only RAD-140 cycle and shared his experiences with the SARM on Reddit. During this time, he took 10 mg of RAD-140 and was in a moderate caloric surplus.

RAD-140 Stacks

RAD-140 can be combined with other PEDs to maximize its effects. RAD-140 is commonly taken with MK-677 (Ibutamoren), YK-11, LGD-4033 (Ligandrol), or Cardarine (GW-501516). Below you'll learn everything you need to know.

Can you stack MK-677 with RAD-140?

Yes, combining these two substances can lead to significantly improved muscle growth. MK-677 is an orally available growth hormone secretagogue. It causes increased secretion of growth hormones and IGF-1. RAD-140 is a SARM that acts similarly to testosterone on the androgen receptor, which leads to its muscle-building effect. Together, these substances maximize this effect and offer additional benefits.

Advantages: Quicker muscle growth andquick recovery, fat loss, both substances are orally available

Disadvantages: Possible water retention, could reduce testosterone production, may cause insulin resistance

Forms of RAD-140

RAD-140 capsules are the most commonly used form. The SARM can also be purchased as a liquid/solution, where RAD-140 is dropped under the tongue in drop form. RAD-140 as a solution is the most bioavailable form. [R]

RAD-140 Dosage

At what dose is RAD-140 effective?

In a study, young male cynomolgus monkeys were given different doses of RAD140. The goal was to determine at what dose RAD-140 becomes effective. The monkeys were divided into three groups. Group 1 received a daily dose of RAD-140 of 0.01 mg/kg body weight. Group 2: 0.1 mg/kg and Group 3: 1mg/kg. The dose was administered for 28 days. [R]

Results: Body weight increased in all 3 groups. Group 1, which only received 0.01mg/kg, had the smallest increase. At doses of 0.1 mg/kg and 1mg/kg, an average weight gain of over 10% in just 28 days was observed. A dose between these two values seems to be optimal for monkeys. [R]

Does RAD-140 build fat?

In the same study, the effects of RAD140 on muscle and fat tissue were examined. Primate tissue weight was measured using a "DEXA" scan on days -2 and 29. [R]

Results: An increase in muscle tissue was demonstrated in all 3 groups, with the 0.1 mg/kg and 1.0 mg/kg groups showing the greatest muscle gains. There was no consistent effect on absolute fat mass. RAD-140 appears to build almost exclusively muscle tissue and not fat. [R]

The History of RAD-140

The development of the SARM RAD-140 began in 2010 by Radius Health [R][R]. Since then, RAD-140 has been primarily researched as a treatment for muscle wasting and breast cancer in women. The first rat trials were conducted in 2014 at the University of Southern California in Los Angeles. Results indicated that RAD-140 could potentially be safer than conventional testosterone replacement therapy (TRT).

RAD140 clinical trials

From 2017 - 2020, RAD-140 was tested in humans for the first time in a Phase I study. A total of 22 women with breast cancer participated [R]. Numerous articles were written about the results, which demonstrated RAD-140's muscle-building effects.

In 2020, Radius Health transferred the rights of RAD140 to Ellipses Pharma, which has continued research on the SARM since then. As of March 2023, RAD-140 is in a Phase 1/2 clinical trial [R].

Do Bodybuilders use RAD-140?

RAD-140 has long enjoyed significant attention in the bodybuilding community and is often used "off-label." Its popularity likely stems from the fact that RAD-140 can be taken orally and is relatively potent.

How to store RAD-140

RAD-140 is available in both liquid and powder form. The same storage conditions apply to both variants:

  • Store RAD-140 in a dark place and avoid exposure to light
  • RAD-140 should be stored at room temperature between 15°C to 30°C (59°F to 86°F)
  • Avoid strong temperature fluctuations
  • Freezing is possible but not optimal
